金雀异黄素和DC疫苗对H22癌细胞抑制作用

Anti-tumor effects of genistein and DC vaccine on mice H22 hepatocarcinoma cells

  • 摘要: 目的 探讨金雀异黄素(GEN)和树突状细胞瘤苗(DC疫苗)对荷瘤小鼠H22癌细胞的免疫抑制作用.方法 采用共聚焦显微镜、流式细胞技术检测H22细胞凋亡的情况,蛋白免疫印迹(Western-blot)检测凋亡相关基因Bax、caspase-3、bcl-2蛋白的表达.结果 共聚焦显微镜观察到GEN DC疫苗诱导凋亡的H22细胞;与1640对照组凋亡率(3.019±0.327)比较,GEN DC组总凋亡率(14.488±1.660)明显增高;GEN DC疫苗诱导可上调H22细胞bax、caspase-3基因的表达而下调bcl-2基因表达.结论 金雀异黄素和DC疫苗对肝癌细胞的生长有一定抑制作用.

     

    Abstract: Objective To investigate anti-tumor effects of genistein and dendritic cell(DC) vaccine on mice H22 hepatocarcinoma cells.Methods Confocal microscopy and flowcytometry were used to detect cell apoptosis in vitro.Westernblot was used to detect the expression of apoptosis-associated gene Bax,caspase-3 and bcl-2 in vitro.Results Genistein and DC vaccine could significantly inhibit the growth and induce the apoptosis of H22 cells.Flowcytometry detection showed that the apoptosis rate of genistein and DC treatment H22 cells was significantly higher than that of 1640 control group(14.488±1.660 vs 3.019±0.327).Genistein and DC vaccine could reduce the expression of apoptosis-regulated gene bcl-2 and improve the expression of apoptosis-regulated gene bax,caspase 3.Conclusion Genistein and DC vaccine can suppress the growth of hepatocarcinoma cells.
