Anti-tumor activity of immuned hemolymph extraction from Musca domestica larvae

  • 摘要: 目的 探讨家蝇3龄幼虫血淋巴40%固相萃取组分抗肿瘤作用及机制。方法 应用四甲基偶氮噻唑蓝(MTT)法检测40%固相萃取组分对S180肉瘤细胞株生长的抑制作用;通过体内试验检测40%固相萃取组分对S180小鼠实体瘤生长的抑制率,测定荷瘤小鼠血清总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量变化。结果 在体外,40%固相萃取组分25 μg/mL对S180肉瘤细胞增殖抑制率最高达79%,与阴性对照组比较差异有统计学意义(t=6.40,P<0.05);40%固相萃取组分有抑制S180实体瘤小鼠肿瘤生长的作用,高、中、低剂量组肿瘤生长抑制率分别为46.23%、35.23%、32.13%;与模型对照组(3.19±0.80)、(163.41±22.65)U/mL比较,3个剂量组的血清T-AOC和SOD活性均升高,其中高剂量组小鼠血清T-AOC和SOD活性分别为(4.91±0.46)、(213.19±43.37)U/mL,差异均有统计学意义(tT-AOC=7.12、tSOD=-7.16,P<0.01);3个剂量组的血清MDA含量均降低,其中高剂量组血清MDA含量为(8.85±1.81)nmol/mL,差异有统计学意义(tMDA=6.79,P<0.01)。结论 40%固相萃取组分在体外和体内均具有抑制S180肉瘤细胞生长的作用,清除自由基可能是其体内抗肿瘤机理。

     

    Abstract: Objective To investigate the anti-tumor effect and its mechanism of Musca domestica larvae of three age by solid phase extraction.Methods The in vitro anticancer effect of the hemolymph extraction from Musca domestica with 40% acetonitrile(ACN)in aqueous solution by solid phase extraction was studied with 3(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-terazolium bromide(MTT) assay; tumor inhibition effect of the extraction on S180 sarcoma-bearing mice was observed.Total antioxygen capability(T-AOC),superoxide dismutase(SOD),and malondialdehyde(MDA) were detected.Results At 25 μg/mL concentration,the inhibitory rate of the S180-cells proliferation was increased to 79%.The inhibitory rate of the cell proliferation was different compared with that of the control group.For low,moderate,and high dosage treatment group,the tumor inhibition rates for mice bearing S180 were 32.13%,36.23%,and 45.23%.The activity of T-AOC and SOD was higher and the content of MDA was lower in the extraction treatment group than those in the control group,with significant differences compared with those of the control group(P<0.05,P<0.01).Conclusion The hemolymph extraction from Musca domestica with 40% ACN solid phase extraction has anti-tumor effect on S180 sacoma cells both in vivo and in vitro and the effect is probably related to antioxidation of the extraction.
