Influences of CYP7A1 gene polymorphism on hyperlipidemia in middle-aged and old populations

  • 摘要: 目的 了解胆固醇7α-羟化酶(CYP7A1)基因多态性对中老年脂代谢紊乱的影响。方法 2008年3月—2009年3月对江苏省南京市2 200名常住汉族中老年人(45~75岁)进行实验室检查,从高脂血症患者中随机抽取223例作为血脂异常组,从健康体检者中随机抽取228人作为对照组,检测血清总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C)和CYP7A1基因多态性。结果 CYP7A1-204 A/C多态位点等位基因A、C频率在血脂异常组和正常对照组分别为30.0%、70.0%和30.7%、69.3%,等位基因频率和基因型频率分布均符合Hardy-Weinberg 平衡定律(P>0.05);血脂异常组中AA、AC和CC 3种基因型的TC水平分别为(4.34±1.08)、(5.67±1.02)、(5.52±1.08)mmol/L,AC、CC型与AA型比较差异均有统计学意义(P<0.05);AA型患者的HDL-C和LDL-C水平分别为(0.88±0.28)、(1.65±0.69)mmol/L,均低于AC型(1.15±0.27)和(2.28±0.61) mmol/L和CC型(1.17±0.32)和(2.31±0.80) mmol/L(P<0.05);正常对照组中仅TC水平在CC型(4.94±0.75) mmol/L和AC型(5.48±0.91) mmol/L间差异有统计学意义(t=5.104,P<0.05)。结论 携带CYP7A1基因突变型的个体,发生脂代谢紊乱的风险增高。

     

    Abstract: Objective To assess the influences of cholesterol 7-alpha hydroxylasel(CYP7A1) gene polymorphism on hyperlipidemia in middle-aged and old populations.Methods We recruited 451 individuals(men and women) aged 45 to 75 years in a population-based cross-sectional survey in Nanjing city,and randomly divided them into dyslipidemia group(N=223) and control group(N=228).Medical examinations,including total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),and low density lipoprotein cholesterol(LDL-C)),and CYP7A1 gene polymorphism,were carried out among the participants.Results The frequency of alleles A and C in CYP7A1 204 in dyslipidemia group and normal control group was 30%,70%,and 30.7%,69.3%,respectively.The allele frequency and genotype frequency distributions were consistent with Hardy-Weinberg balance law(P>0.05).Individuals in dyslipidemia group with AA,AC,CC genotypes showed significant differences in TC(P<0.05),while HDL-C and LDL-C levels of the individuals with A genotype were significantly reduced than those with AC,CC genotypes(P<0.05).The TC levels were significantly different between the individuals with CC and AC genotype in the control group(P<0.05).Conclusion The middle-aged and old populations with the CYP7A1 gene mutant may have an increased risk of dyslipidemia.
