锦州市啮齿动物汉坦病毒分子流行病学特征分析

Molecular epidemiology of hantavirus isolated from rodent hosts in Jinzhou city

  • 摘要: 目的 了解辽宁省锦州市啮齿动物中鼠种构成、带毒率、汉坦病毒(HV)流行情况及病毒分型。方法 采用间接免疫荧光法(IFA)检测鼠肺中HV抗原;用逆转录聚合酶链反应(RT-PCR)法扩增阳性样品中的部分M片段;构建系统发生树进行系统发生分析及分型。结果 2008—2013年锦州市出血热疫区捕鼠1 838只,分属2科4属5种,病毒携带率为4.24%(78/1 838);各年之间鼠肺带毒率有统计学意义(χ2=13.579,P=0.019,P<0.05);部分M片段序列分析表明, 11株病毒与汉城病毒(SEOV)核苷酸序列同源性为96.2%~100%,而与汉滩型病毒(HTNV)同源性仅为70.6%~71.2%,系统进化树显示检测阳性的11株SEO起源相同,与BjHD01株病毒的亲缘关系最近。结论 锦州地区HV优势毒株为SEO型,变异较小,基因型别较为稳定,并有明显的地区聚集性。

     

    Abstract: Objective To study the composition of rodent species,hantavirus(HV)infection rate of the rodents,and types of hantaviruses isolated from the rodents in Jinzhou city of Liaoning province.Methods HV antigens in the rat lungs were detected with immunofluorescence assay.Partial M segment sequences were amplified with reverse transcription-polymerase chain reaction(RT-PCR)and then sequenced.The phylogenetic trees were constructed by maximum likeliood method to detect the genetic characteristics of HV.Results From 2008 to 2013,1 838 host rodents of 2 families,4 genera,and 5 species were captured in the areas with hemorrhagic fever epidemics;the average HV infection rate of the rodents was 4.24%and a statistically significant difference in the annual infection rate was observed(χ2=13.579,P<0.05).Partial M segment sequences were recovered using Seoul virus(SEOV)specific primers from 11 positive samples.The nucleotide homology of these M segment sequences ranged from 96.2% to 100% compared with SEOV and the homology ranged from 70.6% to 71.2% compared with Hantaan virus(HTNV).The established phylogenetic trees indicated that all the 11 isolated viruses belonged to SEOV and the strains showed the nearest relationship with BjHD01.Conclusion The dominant HV strain was SEOV in Jinzhou area,with less mutation,stable genetic type,and obvious regional clustering.
