Association between polymorphism of NR3C1 gene and risk factors of cardiovascular diseases

  • 摘要: 目的 探索糖皮质激素受体基因(NR3C1)单核苷酸多态性(SNP)与心血管疾病常见危险因素的相关性,为探索心血管疾病的遗传因素提供科学依据.方法 采用整群抽样方法,抽取 2012年1—4月在北京市首都医科大学宣武医院体检中心进行健康体检的762名体检者进行体格检查和实验室检测,并采用Sequenom MassArray质谱阵列技术对选取的14个标签SNPs及5个功能性SNPs进行基因分型,比较各SNP位点上不同基因型/等位基因之间各研究指标的差异.结果 rs4143247位点GG基因型携带者的体质指数(BMI)、收缩压(SBP)、舒张压(DBP)分别为(25.50±2.54)kg/m2、(122.90±12.57)mm Hg、(79.30±6.70)mm Hg,均高于CC+CG基因型携带者的(23.46±3.00)kg/m2、(112.99±12.00)mm Hg、(74.51±8.32)mm Hg,空腹血糖(GLU)和甘油三酯(TG)分别为(5.06±0.38)和(0.96±0.22)mmol/L,均低于CC+CG基因型携带者的(5.36±0.71)和(1.22±0.90)mmol/L(均P <0.05);rs10052957位点GA+AA基因型携带者的SBP、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)分别为(112.62±12.12)mm Hg、(4.68±0.84)mmol/L、(2.53±0.69)mmol/L,均低于GG基因型携带者的(116.74±11.83)mm Hg、(5.05±0.88)mmol/L、(2.80±0.70)mmol/L(均P <0.001);rs7701443位点AA基因型携带者的BMI、SBP、DBP、TC、LDL-C分别为(24.67±2.36)kg/m2、(118.53±13.65)mm Hg、(76.64±8.15)mm Hg、(4.97±0.86)mmol/L、(2.75±0.72)mmol/L,均高于GG+GA基因型携带者的(23.38±3.05)kg/m2、(112.63±11.77)mm Hg、(74.42±8.30)mm Hg、(4.73±0.56)mmol/L、(2.56±0.69)mmol/L(均P <0.05);rs17209251位点GG基因型携带者的SBP、DBP分别为(123.78±12.39)、(79.22±7.45)mm Hg,均高于AA+AG基因型携带者的(113.00±12.00)、(74.53±8.30)mm Hg,TC、TG分别为(4.54±0.60)、(0.96±0.23)mmol/L,均低于AA+AG基因型携带者的(4.77±0.87)、(1.22±0.90)mmol/L(均P <0.05);rs2963156位点TT基因型携带者的腰臀比(WHR)、GLU、TC、TG、LDL-C分别为(0.79±0.02)、(4.93±0.18)mmol/L、(4.43±0.50)mmol/L、(0.91±0.26)mmol/L、(2.14±0.20)mmol/L,均低于CC+CT基因型携带者的(0.81±0.06)、(5.35±0.71)mmol/L、(4.76±0.87)mmol/L、(1.21±0.89)mmol/L、(2.60±0.70)mmol/L(均P <0.05).结论 NR3C1基因多态性与心血管疾病危险因素之间存在相关性.

     

    Abstract: Objective To explore the association between single nucleotide polymorphism(SNP)of human glucocorticoid receptor(GR)gene(NR3C1)and the risk factors of cardiovascular diseases.Methods Totally 762 healthy people who took physical examination were involved.Medical information and risk factors of cardiovascular diseases were collected.Fourteen tag SNPs and five functional SNPs in GR gene were selected and genotyped.Results For GG homozygous on rs4143247,the carriers had higher levels of body mass index(BMI),systolic blood pressure(SBP),diastolic blood pressure(DBP)and a lower level of plasma glucose than those with genotypes CC+CG(P <0.05).Compared with genotype GG+GA on rs7701443,BMI,waist-to-hip ratio(WHR)and SBP were higher than those with AA homozygous(P <0.05).For genotype GG on rs17209251,the carriers had increased levels of SBP,DBP,total cholesterol and low-density lipoprotein-cholesterol(P <0.05).Conclusion The polymorphism of GR gene is related to the risk factors of cardiovascular diseases.
