哺乳期磺胺间甲氧嘧啶暴露对仔鼠行为影响

Effects of maternal sulfamonomethoxine exposure during lactating on behaviors of offspring mice

  • 摘要: 目的 探讨磺胺间甲氧嘧啶(SMM)哺乳期暴露对青春期小鼠探索和焦虑行为影响.方法 小鼠哺乳期以0(对照组)、10、50、200 mg/kg灌胃染毒,连续21 d,断乳后其仔鼠雌、雄分笼正常喂养至49 d,次日进行旷场、黑白巷和高架十字迷宫实验.结果 与对照组雄鼠比较,低、中剂量SMM组雄鼠旷场实验中央区域时间分别为(40.6±10.0)、(41.0±11.9)s增加,差异有统计学意义(P <0.05),中剂量组雄鼠站立次数(56.5± 8.8)次明显增加,差异有统计学意义(P <0.01);与对照组雌鼠比较,低剂量SMM组雌鼠旷场实验中央区域时间(37.3±9.0)s增加,差异有统计学意义(P <0.05),中剂量SMM组雌鼠的跨越格子数(159.1±26.0)个增加,差异有统计学意义(P <0.05);高剂量SMM组雌鼠黑白巷实验黑端时间(57.8±8.2)s增加,差异有统计学意义(P <0.01).结论 哺乳期SMM暴露可干扰仔鼠的探索和焦虑等行为.

     

    Abstract: Objective To explore the effects of maternal sulfamonomethoxine(SMM) exposure during lactating on exploratory and anxiety-related behaviors of offspring mice in puberty.Methods The lactational mice were orally administered with SMM(0,10,50,200 mg/kg) for 21 days.Then the offspring mice were separated by gender and raised normally until postnatal day 49,and their exploratory and anxiety-related behaviors were detected with open-field,black-white alley and elevated shelf plus maze test the next day.Results Compared with the control,the central region time of the male offspring mice with 10 and 50 mg/kg maternal SMM exposure increased significantly(40.6±10.0 and 41.0±11.9 seconds,P <0.05);the standing times in the open-field test for the male offspring mice with 50 mg/kg maternal SMM exposure increased significantly(56.5±8.8 seconds,P <0.01).For the female offspring mice compared to those of the control,the central region time significantly increased(37.3±9.0 seconds) in 10 mg/kg maternal SMM exposure group(P <0.05);the total number of squares crossed in the open-field test significantly increased(159.1±26.0) in 50 mg/kg SMM group(P <0.01);and the total time spent in the black half test was also significantly increased(57.8±8.2 seconds) in 10 mg/kg SMM group(P <0.05). Conclusion The results indicate that maternal SMM exposure during lactation affects exploratory and dysphoric behaviors of the offsprings mice in puberty.
