人参皂苷Rg1对2型糖尿病大鼠肝损伤保护作用

Protective effect of ginsenoside Rg1 on liver injury in rats with type 2 diabetes

  • 摘要: 目的探讨人参皂苷Rg1对2型糖尿病大鼠肝脏损伤的保护作用及其机制。方法采用高脂饮食和链脲佐菌素结合的方法建立大鼠2型糖尿病模型, 实验设对照组、模型组、人参皂苷Rg1(2、4、8 mg/kg)组;血糖仪测定大鼠血糖水平, 分光光度法测定大鼠血清谷丙转氨酶(ALT)、谷草转氨酶(AST), 超氧化物歧化酶(SOD)、丙二醛含量;苏木素伊红染色观察肝组织形态学变化;酶联免疫吸附法测定血清中肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)水平。结果与对照组比较, 模型组大鼠血清中ALT、AST分别为(123±10.7)、(138±13.8)U/L、TNF-α、IL-6分别为(54±2.8)、(168±12.5)pg/mL和丙二醛(24.8±2.5)nmol/mgprot含量均增加, SOD(65±7.9)U/mgprot活力降低;与模型组比较, 8 mg/kg人参皂苷Rg1组大鼠血清中ALT、AST分别为(58±8.0)、(65±8.9)U/L、TNF-α、IL-6分别为(34±4.1)、(83±10.4)pg/mL和丙二醛(13.8±1.9)nmol/mgprot含量均降低, SOD(105±9.7)U/mgprot活力增加。结论人参皂苷Rg1对2型糖尿病大鼠肝损伤具有一定保护作用, 其机制可能与其抗炎抗氧化作用有关。

     

    Abstract: ObjectiveTo study the protective effect of ginsenoside Rg1 on liver injury induced by type 2 diabetes in rats and to explore its possible mechanism.MethodsRat type 2 diabetes model was induced by high-fat diet and low-dose streptozotocin(HFD+STZ).The experimental rats were divided into a normal group,a model group and three groups with different dose of ginsenoside Rg1(2,4,and 8 mg/kg).Blood glucose was determined with glucometer.The content of alanine transaminase(ALT),asparate aminotransferase(AST),superoxide dismutase(SOD),and malondialdehyde(MDA)were measured with spectrophotometry.Liver pathological changes were observed with hematoxylin eosin(HE)staining.The content of tumor necrosis factor-α(TNF-α),and interleukin-6(IL-6)were determined with enzyme-linked immunosorbent assay(ELISA).ResultsCompared with the control group,the content of ALT(123±10.7 U/L),AST(138±13.8 U/L),TNF-α(54±2.8pg/mL),IL-6(168±12.5 pg/mL),and MDA(24.8±2.5 nmol/mg prot) were increased and the activity of SOD(65±7.9 U/mg prot) was decreased in model group.Compared with the model group,the content of ALT(58±8.0 U/L),AST(65±8.9 U/L),TNF-α(34±4.1 pg/mL),IL-6(83±10.4 pg/mL),and MDA(13.8±1.9 nmol/mg prot) were decreased and the activity of SOD(105±9.7 U/mg prot)was increased in ginsenoside Rg1(8 mg/kg)group.ConclusionGinsenoside Rg1 has protective effect on liver injury induced by type 2 diabetes and the effect is associated with anti-inflammatory and antioxidative effects of ginsenoside Rg1.
