槲皮素对Con A诱导自身免疫性肝损伤作用

Effect of quercetin on Con A-induced autoimmune liver injury

  • 摘要: 目的 建立刀豆蛋白A(Con A)诱导的小鼠肝损伤模型, 观察槲皮素对Con A诱导的小鼠自身免疫性肝损伤的作用。方法 将48只小鼠随机分为6组:对照组、模型组、地塞米松组、槲皮素低、中、高剂量组(100、200、400 mg/kg), 小鼠连续灌胃7 d后尾静脉注射15 mg/kg Con A, 地塞米松于Con A给药前30 min腹腔注射。分别于尾静脉给药后2、6 h取血清, 检测干扰素(TNF-α)、白介素-1β(IL-1β)、IL-6和丙氨酸氨基转移酶(ALT)、天门冬氨酸转移酶(AST)、乳酸脱氢酶(LDH), 取肝、脾, 称重计算脏器系数。结果 与对照组比较, 模型组小鼠血清中ALT、AST、LDH活性分别为(1 214.20±168.03)、(1 079.40±168.05)、(2 748.00±508.98)U/L明显升高, 细胞因子TNF-α、IL-1β和IL-6水平分别为(580.67±157.01)、(231.67±119.4)、(14 077.5±9 699.09)pg/mL明显升高(P<0.05);与模型组比较, 400 mg/kg槲皮素组小鼠血清ALT、AST、LDH活性分别为(204.20±31.62)、(372.60±29.85)、(1 923.40±418.63)U/L均明显降低, 细胞因子TNF-α、IL-1β和IL-6水平分别为(339.67±76.73)、(108.33±21.47)、(7 068.33±3 414.4)pg/mL均明显降低(P<0.05);肝、脾脏器系数无明显变化。结论 槲皮素对自身免疫性肝损伤具有一定保护作用。

     

    Abstract: Objective To construct a model of concanavalin A(Con A)-induced autoimmune liver injury and to observe the effects of quercetin on Con A-induced autoimmune liver injury in mice.Methods Forty-eight mice were assigned randomly into six groups, including normal control group, model group, dexamethasone group and low-, moderate-, and high-dose quercetin groups(100, 200 and 400 mg/kg).Quercetin(or solvent for control group)were gavaged for 7 days continuously, then 15mg/kg Con A(or solvent)was given to the mice intravenously and dexamethasone was injected intraperitoneally 30 minutes before the injection of Con A.Two and six hours after the treatments, serum of the mice was collected to detect tumor necrosis factor alpha(TNF-α), interleukin 1 beta, (IL-1β), interleukin 6(IL-6), alamine aminotransferase( ALT), aspartate aminotransferase(AST), and lactic dehydrogenase(LDH), respectively.Liver and spleen were weighed to compute organ coefficient.Results Compared with the control group, ALT, AST, and LDH in the model group(1 214.20±168.03, 1 079.40±168.05, and 2 748.00±508.98 U/L)were significantly increased(P<0.01).TNF-α, IL-1β, and IL-6 in the model group(580.67±157.01, 231.67±119.4, and 14 077.5±9 699.09 pg/ml)were significantly increased(P<0.01).Compared with the model group, ALT, AST, and LDH in high-dose quercetin group(204.20±31.62, 372.60±29.85, and 1 923.40±418.63U/L)were significantly reduced(P<0.05).TNF-α, IL-1β, and IL-6(339.67±76.73, 108.33±21.47, and 7 068.33±3 414.4 pg/ml)were reduced significantly(P<0.05).Liver, spleen organ coefficients were not significantly changed.Conclusion Quercetin may have protective effect on autoimmune liver injury induced by Con A in mice.
