褐参提取物对H20荷瘤小鼠肿瘤抑制作用

Inhibitory effect of brown ginseng extraction on H20 tumor cells in mice

  • 摘要: 目的 探讨褐参提取物对H20荷瘤小鼠的抑瘤作用及机制。方法 60只昆明雄性小鼠随机分为5组:模型组、环磷酰胺组、低、中、高剂量褐参组;H20造模24 h后灌胃给予受试物,1次/d、连续9 d,测量小鼠体重、瘤重、脾重、胸腺重量、白细胞数、血清白细胞介素2(IL-2)、肿瘤坏死因子-α(TNF-α)和γ干扰素(IFN-γ)水平及超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)活性。结果 与模型组比较,高剂量褐参组小鼠瘤体重量(1.37±0.42)g明显下降,抑瘤率达39.38% (P<0.05);与模型组比较,高剂量褐参组小鼠血清中TNF-α、IL-2、IFN-γ水平分别为(100.73±10.25)、(62.85±9.92)和(209.20±21.59)ng/L及SOD、CAT、GSH-Px酶活性分别为(304.43±39.51)、(116.24±12.77)、(392.46±40.71)U/mL均明显升高,差异有统计学意义(P<0.05)。结论 褐参提取物对荷瘤小鼠肿瘤具有抑制作用,其机制可能与提高荷瘤小鼠免疫功能和抗氧化能力有关。

     

    Abstract: Objective To investigate tumor suppression effect and its mechanism of brown ginseng extraction in H20 tumor-bearing mice.Methods Sixty healthy male Kunming mice were randomly divided into 5 groups:saline group,cyclophosphamide group,low-,medium-,and high-dose brown ginseng treatment groups.Twenty-four hours after the establishment of H20 tumor model,the mice of treatment groups were gavaged with brown ginseng extraction of different doses one time a day consecutively for 9 days.At the end of the treatment,the weight of body,tumor,spleen,and thymus gland,the count of white blood cell(WBC),the content of tumor necrosis factor-α(TNF-α),interleukin-2(IL-2),and interferon-γ(IFN-γ),the activity of superoxide dismutase(SOD),catalase(CAT),and glutathione peroxidase(GSH-Px)were determined for all the mice.Results Compared with that of the saline group,the tumor weight(1.37±0.42 g)decreased significantly,with a tumor inhibitory rate of 39.38% in the high-dose brown ginseng group(P<0.05).Meanwhile,significant increases in spleen index,WBC count,content of TNF-α(100.73±10.25 ng/L),IL-2(62.85±9.92 ng/L),and IFN-γ(209.20±21.59 ng/L),activity of SOD (304.43±39.51 U/mL),CAT(116.24±12.77 U/mL),and GSH-Px(392.46±40.71 U/mL)were observed in the mice of high-dose brown ginseng group.Conclusion Brown ginseng extraction has inhibitory effect on H20 tumor cells in mice and the effect may be related to the extraction-induced increases in immune function and antioxidation capacity of the mice.
