铅暴露对大鼠海马组织APP和tau蛋白表达影响

Effect of long-term lead exposure on amyloid precursor protein and tau gene expressions in rat hippocampus

  • 摘要: 目的 探讨长期铅暴露对大鼠海马组织淀粉样前体蛋白(APP)和tau基因、蛋白表达影响。方法 将SPF级雌性SD大鼠随机分为3组,分别为对照组(去离子水),低、高剂量铅组(乙酸铅800、1 500 mg/L),所生雄性仔鼠分别继续饮用去离子水(对照组),300 mg/L(低剂量铅组),900 mg/L(高剂量铅组)乙酸铅溶液,分别与生后3周(PNW3)、中年期(PNW41)、老年期(PNW70)测定雄性仔鼠血、海马组织中铅含量;采用荧光定量PCR(qPCR)和蛋白印迹(WB)检测APP和tau mRNA及蛋白表达水平。结果 与对照组比较,各年龄染铅组雄性仔鼠血铅、海马铅含量明显增加(P < 0.05);与对照组比较,高剂量铅组PNW70雄性仔鼠海马APP mRNA和蛋白表达水平分别为(1.398±0.827)、(1.454±0.190)均明显上升(P < 0.05);与对照组比较,低、高剂量铅组PNW3 雄性仔鼠海马tau蛋白表达水平分别为(4.850±1.257)、(5.557±1.880)明显上升(P < 0.01);对照组比较,高剂量铅组PNW70雄性仔鼠海马磷酸化tau蛋白表达水平(3.225±1.098)升高(P < 0.05)。结论 长期铅暴露可导致断乳期和老年期大鼠海马组织APP和tau基因与蛋白表达上调。

     

    Abstract: Objective To investigate changes in amyloid precursor protein (APP) and tau gene expression in hippocampus of rat with long-term lead exposure.Methods Female and male (at the ratio of 2:1) specific-pathogen free Sprague-Dawley rats were randomly divided into three groups:control group supplied with distilled water,low lead exposure group (LLG) with drinking water containing lead acetate of 800 mg/L,and high lead exposure group (HLG) with drinking water containing lead acetate of 1 000 mg/L.Then the male offsprings were continuously exposed to drinking water containing lead acetate of 0 mg/L (control group),300 mg/L (LLG),and 900 mg/L (HLG)mg/L and lead content in blood and hippocampus tissue of the offsprings were measured at postnatal week 3 (PNW3),postnatal week 41 (PNW41) and postnatal week 70 (PNW70);the mRNA and protein expressions of tau and APP in hippocampus of the offsprings were determined simultaneously with real-time quantitative PCR(qPCR)and Western blot.Results Compared with those of the control group,the lead content of blood and hippocampus were significantly increased in lead-exposed male offsprings (P < 0.05).The expression of APP mRNA(1.398±0.827) and protein (1.454±0.190) were significantly up-regulated in PNW70 male hippocampus of offsprings in HLG (P < 0.05).Compared with those of the control group,the expression of tau protein of the offsprings in LLG (4.850±1.257) and HLG (5.557±1.880)were significantly increased at PNW3 (P < 0.01),and the expression of phosphorylated tau protein (3.225±1.098) increased in male offspring's hippocampus of HLG at PNW70 (P < 0.05).Conclusion Long-term lead exposure induces up-regulations of mRNA and protein of APP and tau in hippocampus of rats at PNW3 and PNW70.
