纳米四氧化三铁对大鼠肠道内短链脂肪酸影响

Influence of nano-Fe3O4 on short-chain fatty acids in intestinal tract in rats

  • 摘要: 目的 探讨四氧化三铁纳米(nano-Fe3O4)颗粒对大鼠肠道内短链脂肪酸影响。方法 60只(雌雄各半)SD大鼠随机分为3组,分别为5%纤维素钠对照组、低、高剂量nano-Fe3O4组,每组20只,雌雄各半,每日灌胃给予不同剂量nano-Fe3O4,连续4周,每周以逼迫法采集新鲜粪便样品,采用气相色谱法测定大鼠肠道内的短链脂肪酸含量。结果 染毒第4周时,与对照组比较,低、高剂量nano-Fe3O4组雌性大鼠肠道内丁酸含量分别为(0.05 ± 0.02)、(0.06 ± 0.03) μg/μL明显下降(P < 0.05),低、高剂量nano-Fe3O4组雄性大鼠肠道内乙酸含量分别为(1.31 ± 0.16)、(1.54 ± 0.22) μg/μL明显下降(P < 0.05),高剂量nano-Fe3O4组雄性大鼠肠道内丁酸含量(0.04 ± 0.02) μg/μL下降,差异有统计学意义(P < 0.05);染毒组大鼠部分肠上皮细胞出现空泡变性,杯状细胞排列疏松,柱状吸收细胞减少。结论 此实验条件下nano-Fe3O4颗粒可能对大鼠肠道内短链脂肪酸含量产生影响。

     

    Abstract: Objective To explore the influence of nano-Fe3O4 on short-chain fatty acids in intestinal tract of Sprague-Dawley(SD) rats.Methods Sixty SD rats(half male,half female) were randomly divided into 3 groups:5% cellulose sodium control group,low- and high-dose groups with oral gavage of nanoparticle for 4 weeks.The fresh feces samples of the rats were collected periodically and short-chain fatty acids in the samples were determined with gas chromatography.Results Compared with those of the control group,the concentration of butyric acid for the female rats decreased significantly in low- and high-dose groups (0.05±0.02 and 0.06±0.03 μg/μL)(P < 0.05);the concentration of acetic acid in the male rats of low- and high-dose groups decreased significantly (1.31±0.16 and 1.54±0.22 μg/μl)(P < 0.05);and the concentration of butyric acid (0.04±0.02 μg/μl) in the male rats of high-dose group decreased significantly (P < 0.05) by the end of the treatments.The results of pathological observation revealed vacuoles degeneration in intestinal epithelial cells,loose arrangement in goblet cells,and the number of absorptive cells decreased in the colon of rats in exposed group.Conclusion The results suggeste that nano-Fe3O4 may affect short-chain fatty acids in intestinal tract of SD rats.
