PI3K/AKT/mTOR信号通路相关蛋白在胃癌中表达

Expression of proteins associated with PI3K/AKT/mTOR signaling pathway in gastric cancer tissues

  • 摘要: 目的 探讨PI3K/AKT/mTOR信号转导通路中PTEN、p-AKT和p-mTOR蛋白在胃癌中的表达及其与临床病理特征的关系。方法 应用免疫组化法检测胃癌、癌旁和正常胃黏膜组织中PTEN、p-AKT和p-mTOR蛋白的表达情况,并分析其与病理参数之间的关系。结果 PTEN在胃癌中的表达率为53.03%,在癌旁与正常胃黏膜中的表达率为84.85%和74.36%,p-AKT在胃癌中的表达率为56.06%,在癌旁与正常胃黏膜中的表达率为16.67%和2.56%,p-mTOR在胃癌中的表达率为90.91%,在癌旁和正常胃黏膜中的表达率分别为71.21%和5.13%,3组间差异均有统计学意义(P<0.001)。PTEN的阳性表达与患者年龄、性别、肿瘤的直径、分化程度、浸润深度、淋巴结转移以及TNM分期均无关(P>0.05)。p-AKT在胃癌中的表达与患者年龄、性别和肿瘤直径无关(P>0.05),而与肿瘤分化程度、浸润深度、淋巴结转移和TNM分期有关(P<0.05)。p-mTOR在胃癌中的表达仅与肿瘤浸润深度有关(P<0.05),与患者年龄、性别、肿瘤的直径、分化程度、淋巴结转移以及TNM分期均无关(P>0.05)。PTEN蛋白与p-AKT蛋白阳性表达率之间存在负相关关系且具有统计学意义(P<0.05),p-mTOR蛋白与PTEN蛋白以及p-mTOR蛋白与p-AKT蛋白表达阳性率之间均无明显相关(P>0.05)。结论 PTEN在胃癌组织中低表达,p-AKT和p-mTOR在胃癌组织中高表达,PI3K/AKT/mTOR信号转导通路可能参与了胃癌的发生与发展。

     

    Abstract: Objective To study expressions of phosphatase and tensin homologue deleted on chromosome 10 (PTEN),phosphorylated protein kinase B (p-AKT),and phosphorylated mammalian target of rapamycin (p-mTOR)in gastric cancer tissues and their relevance with clinicopathological features of gastric cancer.Methods Immunohistochemistry technology was used to detect expressions of PTEN,p-AKT and p-mTOR in gastric carcinoma tissues,tumor-adjacent tissues and normal gastric mucous tissues and their relationships with pathological parameters were analyzed.Results The PTEN level in gastric carcinoma tissues was significantly lower than that in tumor-adjacent and normal gastric mucous tissues;while the levels of p-AKT and p-mTOR were significantly higher than those in tumor-adjacent and normal gastric mucous tissues (P<0.05 for all).The expression of PTEN was irrelevant to clinicopathological features.The expression of p-AKT was associated with the differentiation degree of tumors,infiltration depth,lymph node metastasis and tumor-node-metastasis(TNM) staging (all P<0.05);however,there were no relationships with age,gender and tumor sizes (P>0.05 for all).The expression of p-mTOR in gastric carcinoma tissues showed no relationship with the clinical histopathological features but the depth of invasion.The expression of PTEN was negatively correlated to the p-AKT expression (P<0.05);but there was no significant correlation between p-mTOR protein and PTEN protein and p-AKT protein expression (both P>0.05).Conclusion The PTEN expression is down-regulated and the expression of p-AKT and p-mTOR are up-regulated in gastric cancer tissues,suggesting that PI3K/AKT/mTOR signaling pathway may be involved in the occurrence and development of gastric cancer.
