Study on Genotype of Hantavirus Strains Isolated From Different Areas In Hubei Province

  • Objective To analyse the M genomic segment of 30 strains of Hantavirus(HV)isolated from different areas in Hubei province,in order to understand the genetic character istics and the genomic function of HV in Hubei proince.Methods According to Genebank sofeware of HV,the type-specific primers were designed correspanding to the conser ved sequences in the G1 region of M genomic segment of HV.RT-heminested PCR and restriction mapping methods were applied to type and to digest 30 strains of HV isolated from different areas in Hubei province.Results Thirty strains isolated from different areas in Hubei province were typed as 21 HTNV and 9 SEOV,respectively.The restriction mappings of 21 isolates digested by HinfⅢ and HaeⅢ were consistent with those of 76/118 standard strain,but it was different from those of H- 114 str ain,of which digested by both HaeⅢ in only one segment in Wuhan and Honghu City and HinfI in three segments in YingCheng、Jianx ia、Huangpi and M acheng City respectively.Then all HTNV were digested by HincⅢ except the above strain in Yingoheng City.In addition,9 isolates were digested by HinfⅢ in two sements or EcoRI in two segments while not being digested by HaeⅢ,which were not consistent with those of R22 strain.Conclusion Hubei province was an mixed epidmic areas of HFRS.The M genomic segment of SEOV was stable,but that of HTNV no stablility.Distincthantaviruses could coexist in either different or the same geographic orecological zores in Hubei province.
