Effect of oxidative stress induced by nickel sulfate on testis cell of rat

  •   Objective   To study the ox idative stress effect of nickel sulfate(NiSO4)on rats.testis, and to provide the laboratory theoretical evidence forpreventing and curingof male reproductive tox icity.
      Methods   32 male Wistarrats with sex ual maturation were divided intofourgroups randomly, and three groups of rats were administrted intraperitoneally with nickel sulfate daily at doses of 1.0, 2.0, 4.0 mg/kgrespectively for30 days, but the control group was administrated with saline in same volume according to the rats.weight.The testis homogenates were prepared to detect the levels of lipid perox idation(LPO), total antioxide capacity(T-AOC), anti-reactive oxygen species(anti-ROS), glutathioe periox idase(GSH-Px)in spectro pho to metric assay.
      Results   The organ coefficient of testis exposured to NiSO4 had no change compared with the control group(P > 0.05).The LPOcontents in testis of the 2.0, 4.0 mg/kg groups were higherthan that in the control group(P < 0.05);The activities of anti-ROSand GSH-Px of the 2.0, 4.0 mg/kg groups were lowerthan those in the control group(P < 0.01).The T-AOClevels of the exposure groups were decreased compared with the control group(P < 0.01).
      Conclusion   The male reproductive tox icity is related to the enhancement of oxidative stress effectinduced by nickel sulfate.
