Adjusting effect of diallyl trisulfide on cytokines secreted by kupffer cells

  • ObjectiveTo study the protecting effect of diallyl trisulfide on rat liver from chronic injury caused by high-fat diet, and adjusting effect of diallyl trisufide on cytokines secreted by Kupffer cell(KCs)in response to endotoxin(Lps) stimulation.MethodsFemale Wistar rats were randomly divided into three groups, the chronic injury model induced by high-fat diet, and dug gery group which were given diallyl trisulfide. After 10 weeks, rat were sacrificed, blood sample was collected from abdominal artery for testing ALT、IL-6、IL-10、TNF-α.Liver was made paraffin sections and the rest mice were used as KCs isolated to assess function of KCs secretion after LPS stimulation.ResultsThe levels of ALT、IL-6、TNF-α in model rat obviously rose(P<0.05).The diallylf trisulfide could reduce ALT activity and decreased the level of IL-6(P<0.05).The contents of ALT、IL-6 and TNF-α in supernatant were exactly similar to the results mentioned above after the cultured KCs were simulated by Lps,but the levels of IL-10 were higher than thatin the druggery group(P<0.05).ConclusionDiallyl trisulfide could protect rat liver from chronic injury induced by high-fat diet and adjustcy to kines secreted by Kupffer cells.
