Analysis on cytotoxicity of organic pollutants in river of Chongqing City

  • ObjectiveTo investigate the org anic pollution status of Jialing River and Yangtze Riverin Chongqing section,and to evaluate the effects of organic pollutants in river water on public health.MethodExtracted organic pollutants using solid phase extraction,dissolved the organic pollutants in dimenthy sulphoxide(DMSO),and the extracts were exposed to the H4IIE cell,used 3-(4,5-dimethy lthiazol-2-yl)2,5-diphenylteyr a-zolium bromide(MTT)assay to test the proliferation inhibition of the extracts on the H4IIE cell,and fluorometry was used to test the Etho xy-Resorufin-O-Deethylase(EROD)inducement of the extracts on the H4IIEcell.ResultAfter exposed to the extracts,the proliferation of H4IIE cell decreased according to the exposure time and dose.The 48h and 72h treatments of 200×,and 250×concentrated samples exhibited significant reductions(P<0.05,vs control)to the cell proliferation.The EROD tests showed that the EC50 value for 2,3,7,8-tetrochlorodibenzo-p-dioxins(TCDD)was 4.4 pg/ml,and the toxic equivalency(TEQ)values of each sample corresponding to 2,3,7,8-TCDD were 2004.08 Daxigou:2.2×10-4 pg/L;2004.08 Cuntan:0.9×10-4 pg/L;2005.01:Daxigou:13.3×10-4 pg/L;2005.01 Cuntan:3.7×10-4 pg/L respectively.ConclusionThe toxicity of o rganic pollutants to the H4IIE cell showed time and dose dependence;all of the samples could induce the H4IIE cell EROD activity;the toxicity of low water period samples was higher than that of the high water period samples at the same sampling site.
