Study on excitatory amino acids in PFOS-exposed rats by HPLC

  • ObjectiveTo explore the time-response relationship between central neural excitatory amino acids(EAAs) and PFOS.MethodsWister rats were randomly divided into l control group and 4 duration2,4,8,24 h after 25mg/(kg·bw)exposuregroups.Wister rats were exposed once orally.The controls were treated with 2% tween-80.The contents of glutamate(Glu),aspartate(Asp),glycine(Gly)and gamma-aminobutyric acid(GABA)in brain tissues were determined by high performance liquid chromatography(HPLC).ResultsGlu content at 2,4,8 h were(0.89±0.162),(0.75±0.276), (0.84±0.542)μmol/(g·protein),and there was significant difference compared with corresponding control group(1.30±0.063)μmol/(g·protein)(P<0.05);Asp did not show evident changes;Gly content at 8,24h were(1.24±0.352), (1.09±0.215)μmol/(g·protein),and there was significant difference compared with corresponding control group(1.54±0.120)μmol/(g·protein)(P<0.05).GABA content at 24h were(1.68±0.188)μmol/(g·protein),and there was significant difference compared with corresponding control group(2.03±0.370)μmol/(g·protein)(P<0.05).ConclusionEAAs appears to be related with the development of inhibited activity and then excitatory symptom by the low dose of PFOS exposed.
