Study on effect of PFOS exposure on excitatory amino acids in rats

Abstract
ObjectiveTo dicuss the dose-effect of perfluorooctane sulfonate on excitatory amino acids(EAAs)in rat central nervus system.MethodsWistar rats were treated once by gastric perfusion method with 215,25,50 mg/kg PFOS and the controls received same valume vehicle(2% tween-80).After 24 hours of the treatment,the rats in all groups were sacrificed.The contents of glutamate(Glu),aspartate(Asp),glycine(Gly)and gamma-aminobutyric acid(GABA)in brain tissues were determined by high performance liquid chromatography(HPLC).ResultsAsp content in 12.5,50 mg/kg group were(1.24±0.109),(1.11±0.452)μmol/g,and there was significant difference compared with control group(1.51±0.076)μmol/g(P<0.05).Glu did not show evident changes.Gly contents in 12.5,25,50mg/kg group were(0.82± 0.201),(1.09±0.215),(1.12±0.395)μmol/g and there was significant difference compared with control group(1.54±0.120)μmol/g(P<0.05).GABA contents in 12.5,25 mg/kg group were(1.52±0.187),(1.68±0.188)μmol/g,and there was significant difference compared with control group(2.03±0.370)μmol/g(P<0.05).ConclusionEAAs appears to be related with the development of inhibited activity and then excitatory symptom of central nervus system after low dose PFOS exposure.
