DNA damage of lymphocytes caused by chloropropanol in mice

  • Objective To observe the damage of lymphocyte DNA induced by 1,3-dichloro-2-propanol(1,3 DC P) and 2-methy l-3-3-3 trichloro-1,2-propanedid(3-MCPD)in male mice with comet a ssay.Methods Fifty-six KM mice were divided randomly intofour groups:control group,low dose group(10 mg/kg),medium dose group(20mg/kg),and high dose group(40 m μg/kg).Peripheral blood of the mice was extracted after exposed for seven days.Comet a ssay was made with peripheral blood lymphocytes.Results The DNA dam age of lymphocytes in mice of the 1,3 DCP group was significantly higher than that of the control group.The rate of DNA damage were 22.90%,46.75% and 73.80%,respectively.Morevoer,DNA migration in high 1,3 DCP group was significantly higher than that of the low 1,3 DCP group.DNA migration was 8.056±2.291,9.378±3.658,and 2.3764±0.315,respectively.After the contamination,no significant difierence of DNA damage in mice were observed among the 3 MCPD groups(P>0.05).Conclusion 1,3 DCP has damage effect on DNA of peripheral bloodlymphocyte in male mice.The expermient of 3-M CPD does not show significant impacton DNA of lymphocytes in male mice.
