Study of HIF-1α and Caspase-3 on lung anoxic injury caused by gas explosion in rats

  •   Objective   To explore the dynamic changing regularity of HIF-1α mRNA and its production, and to study the roles they played in the consequent damage in lung anoxic injury caused by gas explosion.
      Methods   animal model of gas explosion injury was created.The lung features were analyzed after fire gas explosion in 30 min, 12h, 24 h.Semi-quantitative RT-PCR was used to measure HIF-1α mRNA.Immunohistochem of HIF-1 and Caspase-3 in the lung tissue was measured.
      Results   The expression of HIF-1α mRN A and its protein rised obviously and peaked at 2 hours.It was higher than that at 30 min and 48 hours.There was not expression in control group(P < 0.01).The expression of Caspase-3 rised obviously and peaked at 2 hours.
      Conclusion   HI F-1α played an important role in lung anoxic injury and the consequent damage caused by gas explosion, and it correlated with the injuring severity.The reduce of over expression of Caspase-3 and HIF-1αcould alleviate the damage and inflammation reaction in lungs of rats.
