Objective A modified method based on high performance liquid chromatography was developed for the determination of aceclofenac in human plasma.
Methods Aceclofenac was extracted from plasma with diethylether.The analytical column was packetd with Hypersil-C18(4.6 mm×250 mm, 10 L).The mobile phase was methanol-water-glacial acetic acid(73B30B0.5, V/V)and the flow rate was 1.5 ml/min.The UV detection wavelength was 276 nm.
Results The calibration curve was liner in the range from 0.1 to 100μg/ml(r=0.996 1), with determination limit of 0.1 μg/ml and the average recovery of(101.17±1.17)%.The intra-day RSD and inter-day RSD were(6.04±2.45)%and(5.01±1.39)%, respectively.
Conclusion The method was simple, rapid, accurate and can be used to determine the aeclofenac concentration in plasma and for pharmacokinetics study.
