Study on rat fetuses talipes equinovarus model induced by all-trans retinoic acid

  •   Objective   To establish rat fetuses talipes equinovarus model induced by all-trans retinoic acid and investigate the optimum dosage of all-trans retinoic acid.
      Methods   Wistar rats were distributed randomly into a control group and 4 assay groups.On day 10 of pregnancy, variable dose of 120, 130, 135 and 140 mg/(kg°bw)all-trans retinoic acid suspended in miner al oil was given intragestrically to the rats of each assay group and equivalent volume of mineral oil of the 140 mg/(kg°bw)group was given to the control rats.Fetuses were delivered and examined under stereomicroscope on day 21 of pregnant.The hindlimbs were removed, processed, sliced up as serial sections in three planes of foot, analysed under a light microscope.
      Results   Deformity of talipes equinovarus and other external malfomlations were induced by all-trans retinoic acid on Wistar rat fetuses.The incidence of simplex deformity of talipes equinovar us was the highest in the 135 mg/(kg°bw)assay group.
      Conclusion   135 mg/(kg°bw)all-trans retinoic acid was the optimal dosage for inducing deformity of talipes equinovarus.
