Effect of nonylphenol (NP) on mRNA and protein expression of in ovarian cancer

  •   Objective   To explore the effects of nonylphenol(NP)on mRNA and protein expression of c-myc and p53 in ovarian cancer PEO4 cells.
      Methods   Human ovarian cancer PEO4 cells were grown in DMEM medium.MTT assay was employed to investigate cell proliferation.RT-PCR and immunohistochemistry was performed to determine mRNA and protein expression of c-myc and p53.
      Results   Within the range of(8-96)×10-7 mol/L, NP stimulated proliferation in PEO4 cells with a dose-dependent and time-dependent fashion.RT-PCR and immunohistochemistry analyses revealed that treatment with 32×10-7 mol/L for 72 h resulted in increased mRNA and protein expression of c-myc and p53 with 2-3 fold.
      Conclusion   NP possessed estrogenic activity and could markedly stimulate proliferation in estrogen responsive ovarian cancer PEO4 cells.Up-regulation of c-myc mRNA and protein expression was suggested to be involved in this biological effect.
