Abstract(460) HTML (75) PDF 1195KB(53)
Abstract:
Objective To study the role of cholesterol in the envelope of human herpesvirus-6(HHV-6)when they enter host cells. Methods HHV-6A GS strains were treated with different concentrations of methyl-β-cyclodextrin (MβCD).The treated virus was subjected to ultracentrifugation through a 20% sucrose cushion to remove MβCD and purified virons, then HSB-2 and J urkat cells were infected by treated HHV-6A respectively. The roles of HHV-6A binding host cells, induced cell-fusion and the ability of entry were examined by using IFA,flow cytometry and Western blotting post HHV-6A cholesterol depeletion. Results When cholesterol was removed from HHV-6 virions with 10mmol/L MβCD,the expression of IE1 was not detected, the infectivity was abolished, but it could be partially rescued by the addition of exogenous cholesterol.50 mmol/L may partially restored the infectivity of HHV-6 treated with 2.5 mmol/L MβCD.Compared with untreated with MβCD,HHV-6 binding was affected slightly by MβCD treatment. In contrast, envelope cholesterol depletion markedly affected HHV-6 infectivity and HHV-6-induced cell fusion. Conclusion Cholesterol present in the HHV-6 envelope plays a prominent role in the fusion process and is a key component in viral entry.

Abstract:
Objective To determine the relationship between symptoms and health related quality of life(HRQOL)in patients with hypertension. Methods A cross sectional study was used to survey 644 patients with hypertension using the questionnaire.Spearman rank correlation method was used to test the relationship between symptom and every scale of SF-36 and stepwise multivariate regression method was used to test the impact of different risk factors on quality of life. Results Symptom counts and symptom distress scores were significantly correlated with eight scales of SF-36.In multivariate models, higher symptom counts and sympto mdistress scores were strongly associated with lower HRQOL scores among both patients taking antihy pertensive drug and those without drug. However, they were greater in magnitude than any other predictors, including age, blood pressure among patients taking drug. Conclusion Symptoms have great impact on HRQOL.Symptom monitoring and management are central to the improvement of patients' HRQOL.

Abstract(642) HTML (149) PDF 338KB(170)
Abstract:
Objective To investigate the diagnostic value of serum total immunoglobin E(IgE)test in respiratory allergies. Methods 475 patients registered to department of respirology or allergy clinic in the hospital were evaluated for serum total IgE.The level of specific IgE(SIgE)to common inhalant allergens was also detected. Results The TIgE values in these allergic patients were associated with a non-normal distribution pattern.Compared with young and older adult groups,0~14 years-old group showed higher TIgE level with markedly statistical significance(P=0.001).Within the normal reference range(<1.3 KU/L),38% of the subjects were found to be clinically symptomatic, and some of them concurrently demonstrated mite-specific IgE positive. The level of SIgE increased with higher TIgE.Conclusion Serum TIgE test may not be effective to eliminate the involvement of allergies to common inhalant allergens in symptomatic patients, whereas higher TIgE level may be linked to higher risk of allergies.Normal reference value of TIgE originated from normal populations should not be extrapolated to identify individuals with positive or negative results from allergic test due to a wide range of overlapping between the two.
