Abstract(1089) HTML (246) PDF 564KB(30)
Abstract:
Objective   To study aberrant DNAmethylation potentially resultingin the changes of the anti -7, 8, -dihydrodiol-9, 10-epoxide benzo[a]pyrene(anti-BPDE)and nickel sulfide(NiS)transformed Human bronchial epithelia(16 HBE)as a possible epigenetic mechanism forcarcinogenesis.   Methods   The product DNAisolated from five kinds of 16 HBEwas analyzed foraberrant methylation using PCRbased technique-methylation sensitive restriction fingerprinting technique(MSRF).Several DNAfragments differentially methylated in the transformed cells found by MSRFwere ligated to pMD18 TVectorand transformed into bacteria. The plasmid DNAwere sequenced and compared with data in GenBank by BLASTN.   Results   DNA hypermethylation was indentified in the NiStransformed 16 HBE, and it was found that one of the DNAfragments homologized(99%)with ANKRD11 which encoded the susceptibility protein of nanopbaryngeal carcinoma and anotherhomologized(99%)with bomeo box A3.   Conclusion   Anti-BPDEinduced 16 HBEdid not showaberrant genome Cp Gmethylation, which suggestes that aberrant methylation of genes may nothappen foranti-BPDE-induced cells transformation and carcinogenesis.DNA hypermethylation is known to resultin gene silencing, and it appears that hypermethylation of gene may represent a possible epigenetic mechanism forNiSinduced cells transformation and carcinogenesis.

Abstract:
Objective   To establish SYBRGreen I real-time ramification amplification method(RAM)fordetection of Escherichia coli O157:H7.   Methods   The bacteria were diluted in 100-fold serial with saline from 107 to 10 bacteria/Ll, and detected by rea-l time RAMfordetermining the sensitivity.Clinical isolates were detected forfurtherstudy.   Results   The lowest numberof targets detected by rea-l time RAMwas 10 bacteria/Ll, and the time of appearance of detectable signal was depended on the target concentration.Three clinical isolates of E.coli O157:H7 were positive forshig a tox in gene, while nonpathogenic E.coli were negative.   Conclusion   SYBRGreen Ireal-time RAMis rapid, accurate, simple, real-time environ-mental protecting, and could be another DNA amplification method to detect E.coli O157:H7.

Abstract:
Objective   To establish an economical and effective screening method forundiagnosed diabetes in Shanghai.   Methods   Based on an epidemiological survey of diabetes mellitus in the community populalion in Shanghai, targe-tpopulation was selected to develop risk factors scoring method and logistic regression model. The targe-tpopulation was divided training group and testing group.Logistic regression analysis on the o riginal data of training group was carried out to screen the main risk factors of diabetes mellitus. The score of the variables was determined based on the 10* Bvalue and the aggregate score and aggregate incldence were used to predict the risk of undiagnosed diabetes. The validity and reliability of two methods were validated and compared based on the original data of testing group.   Results   When the threshold value was set up at 18, both sensitivity and specificity of risk factors scoring method foridentifying undiagnosed diabetes were 61.33% and 80.40% respectively. When the threshold value was set up at 0.05, both sensitivity and specificity of logistic regression model were foridentifying undiagnosed diabetes were 67.35% and 76.19% respectively.   Conclusion   The screeningeffect between risk factors scoring method and logistic regression model were no significant. The risk factors scoring method could serve as an effecti and useful tool formass screeningof diabetes mellitus.

Abstract:
Objective   To study the infective factors of the diabetic nephropathy class.   Methods   The application of SPSS12.0 statistics software with the method of ordinal regression.   Results   The degree of diabetic nephropathy was related to urinary microalbumine(Alb)and blood creatinine. When the Alb increased an unit without otherinfluences, the probability of the degree of the diabetic nephropathy to increase one rank ormore was 2.43%;meanwhile, when the blood creatinine increase one unit without the effect of the Alb, the probability of the degree of the diabetic nephropathy to increase one rank or more was 2.22%.   Conclusion   Ordinal regression can not only show the infective factors of the diabetic nephropathy, but also can make quantitative analysis as well as individual prognostication.

Abstract:
Objective   To investigate the possible association of MICAgene polymorphism, especially allele and phenotype of allele 5.1(A5.1), with the risk of gastric cancerand evaluate the possible association of HPpositive ornegative with the gene polymorphism.   Methods   MICApolymorphism was analyzed in 100 gastric cancerpatients and 220 were randomly selected unrelated controls in Jiangsu province, China by usinglocus-specific prime polymerase chain reactions(PCR), followed by electrophoresis of the PCRproducts in denaturing polyacrylamide gels and subsequent detection by silverstaining.HPwas detected by ureCPCR.   Results   The phenotype frequency of allele A5.1 of MICAin subjects with gastric cancerwas significantly higherthan thatin controls, as was the frequency of allele.Simultaneously, the phenotype frequency of allele A5 of MICAin subjects with gastric cancerwas significantly lowerthan thatin controls, as was the frequency of allele.   Conclusion   Allele A5.1 in MICAmight conferthe risk of gastric cancer, and allele A5 seems to be one of protective factors against development of gastric cancer.

Abstract:
Objective   To investigate Ras-Raf-ERK 1/2 mediated functions of basic fibroblast growth factor(bFGF) on proliferation and apoptosis in ovarian cancercell CAOV 3 and to study the relationship between signal transduction mechanism of bFGFand the development of ovarian cancer.   Methods   Aftertreated with bFGFand PD98059, the inhibitorof mitogen-activ ated protein kinase(MEK 1), proliferation rate, apoptosis rate, ext racelluarsignal-regulated protein kinase(ERK1/2)activity of CAOV 3 cell were studied by means of methyl thiazoly l tetrazolian(MT T)assay, FACScan flowcy tometer, Western blot respectively.   Results   bFGFcould dose dependently improve the proliferation of CAOV 3 cell significantly.Treated cells with 75 ng/ml bFGF, the pro liferation rate increased to 140%: bFGFdecreased the apoptosis rate induced by starvation[(33.20±5.32)% ~2.38±3.36)%]; bFGFinduced ERK 1/2 activity. All these effects of bFGFwere inhibited by PD98059.   Conclusion   In ovarian cancercell CAOV3, bFGFincreased pro liferation rate, decreased apoptosis rate, which was mediated by Ras-Raf-ERK1/2 pathway. The bFGFsignal transduction might play important roles in the development of ovarian cancer.

Abstract:
Objective   To explore the hepatocellular DNA damage aftersubacute formaldehyde exposure and the application of normalization as a statistic method to improve the reproducibility of single cell gel electrophoresis(SCGE, comet assay).   Methods   Mice were respectively exposed to different concentrations of gaseous formaldehyde for 14 d, 6 h perday.SCGEwas performed to detect the DNAdamages. And normalization was used to discuss the reproducibility of SCGEassay.   Results   Formaldehyde could induce different kinds of DNAdamages: formaldehyde caused significant DNAstrands breaks at concentrations of 0.5 and 1.0 mg/m3(P < 0.001)while crosslinks were induced at 3.0 mg/m3.   Results   from normalization were in consistent with the formerresults and indicated that DNAdamages were enhanced with the increase of formaldehyde level.   Conclusion   Formaldehyde was both a DNAstrands breaks and crosslinks agent to the hepatocytes in vivo, which may be one of the possible mechanisms of formaldehyde as a carcinogen.Normalization could improve the reproducibility of SCGEassay.
