A murine local lymph node assay: Daicel adenosine triphosphate test on eleven blind samples in BALB/c mice

  •   Objective  To evaluate the allergenicity of 11 blind samples with murine local lymph node assay: Daicel adenosine triphosphate (LLNA: DA) test method and to verify whether the CBA/J mice could be substituted with BALB/c mice in LLNA: DA test recommended by Organization for Economic Co-operation and Development (OECD) guideline.
      Methods  Three dosages were determined for each of the 11 unknown chemical materials with pre-test. Four specific pathogen free (SPF) BALB/c mice of 8 – 12 weeks old were used for each dosage group. One hour after smearing of 1% sodium lauryl sulfates (SLS), the various dosages of bland samples dissolved in acetone (ACE), dimethyl sulfoxide (DMSO), or ACE and olive oil (4 : 1 v/v) were administered to the back of the mice′s ears four times in a week (on the 1st, 2nd, 3rd and 7th day). Specimens of ear tissues and lymph nodes of the mice were collected one day after the last administration. Adenosine triphosphate (ATP) content was measured with ATP fluorescence detector and the stimulation index SI and EC1.8 were determined.
      Results  Compared with those from referral datasets of classic guinea pig test, the consistency rates were 82%, 73% and 91% for the results the tests with 2, 3 and 4 mice; while, between the results of LLNA and LLNA: DA, the consistency rates were 64%, 55%, and 82% for the results the tests with 2, 3 and 4 mice, respectively.
      Conclusion  Compared with both the classic guinea pig experimental database and the LLNA database, the results of the tests with four mice were more sensitive and more accurate, indicating that CBA/J mice could be substituted with BALB/c mice in LLNA: DA test recommended by OECD guideline.
