The Effect of Iron and Ethanol On the Lipid Peroxidation of Liver Tissue

  • To observe the effect of difference content of iron in liver on liver damage induced by ethanol and to investigate the interaction between iron and ethanol in inducing liver damage.Feeding rats a chow diet supplements with carbonyl iron developed experimetal iron-loaded rats.Different iron loaded liver homogenates were cultrured with certain dose of ethanol.The iron content,oxidative and lipid peroxidation induced by ethanol was analyzed.The result show that compared to control rats,the content of iron in liver from rats feeding a chow diet supplement with 2.5%(w/w)carbonyl iron for 2 to 6 weeks increased significantly with time-depend pattern.Lipid peroxidation of normal liver homogenates incubated with difference dose of etnanolwas not significant difference from control.On the other hand,the MDA content in different iron loaded homogenates cultured with certain dose of ethanol(0.5ml)distinctly increased and the GSH content drastically decreased compared to the control.There is a dose-responseds relation between them.The result suggests that the content of iron in liver play a key role in lipid peroxidation induced by ethanol.There is a synergistic hepatotoxicitic effect between ethanol and iron overload through inducing liver tissue lipid peroxidation.
