Study on DNA damage by air pollutants in transport station of chemical dangerous goods

  •   Objective   To explore the DNA damage induced by air pollutants in a transport station of chemical dangerous goods(TSCDG).
      Methods   SOS/umuassay and the ethidium bromide flurescence assay and the single cell gel electrophoresis assay were applied respectively to detect the DNA damage and DNA cross-links and DNA single strand breaks after exposure to the particle extracts of air pollutants in TSCDG.
      Results   The air pollution of TSCDG at asingle dose of 81, 163, 325, 650 g.The results showed that air pollution in TSCDG induced significant SOS response to TA1535/PSK 1002 strain and exhibited obvious dose-response relationships(r=0.994, P<0.01).It had not showed genotoxicity with liver microsomal metabolic activation.The air pollution of TSCDG at a single dose of 1.02, 2.03, 4.06, 8.13 mg/ml.DNA cross-links and DNA single strand breaks in rat type lung cells were found after exposed and the dose-response relationships were existed between formation of DNA cross-links or DNA single strand breaks on rat type lung cells and exposed concentrations of the extracts(rcross-links=0 981, rbreaks=0.987, P<0.01).
      Conclusion   The air pollutants in TSCDG may result in DNA damage.
