Study on time dependent changes of nerve electrophysiology induced by allyl chloride in rats

  •   Objective   To study the time dependent changes of sciatic nerve electrophysiology caused by allyl chloride in wistar rats.
Methods   90 M ale Wistar rats weighting 180~220 g were divided rando mloy into nine groups, i.e.experimental groups for 3, 6, 9, 12 weeks and control group for 0, 3, 6, 9, 12 weeks, each 10 animals. The animals in experimental groups were treated with allyl chloride dissolved in cornoil(200 mg/kg ip 3 days/week)by gavage.Electr ophisical indexes of each group were determined on the corresponding deadline. The indexes included measurements of sciatic nervous conduct velocity, action potential amplitude, latency, wave length, threshold intensity and maxs timulate intesity.
Results   All indexes of electrophysiology were changed gradually in rats treated by allyl chloride. The nervous conduct velocity significantly decreased as compared with the control rats, in 6-week experimental group(P < 0.05), 9-week experimental group(P < 0.01), 12week experimental group(P < 0.01)and the decrease(P < 0.01)accompanied 2, 3, 4 gait scores; amplitude decreased as compared to the control rats(P < 0.05), in 12-week experimental group and the decrease(P < 0.01)accompanied 4 gait score. While the rest indexes were significantly increased(P < 0.05)in 9-week exper imental group, 12-week experimental group and the increase(P < 0.01)accompanied 3, 4 gait scores.
Conclusion   The time dependent changes in electrophysiology of wistar rats sciatic nerves caused by allyl chloride has occured. The nervous conduct velocity may be used as the most sentitive indicator to the early diagnosis for the allyl chloride toxicosis.
