Objective To observe the effects of lead on proliferation of hematopoietic cells in umblical cord blood(UCB).
Methods The concentration of lead was detected in 21 UCBs.After being harvested by centrifugation with Ficoll Hypaque lymphocyte separating medium, the mononuclear cells(MNCs)were cultured in vitro for 15 days and their proliferation was observed periodically.
Results The concentration of lead in UCB was(54.3±1.7)μg/L.The difference of cell density between various lead content was significant.The amplification of MNCs with high lead level was inferior to that with low lead level.The inhibition of lead from proliferation and differentiation of MNCs was resented mainly in the early days during culture and the effect was weakened at the later stage.
Conclusion The effect of lead on proliferation and differentiation of UCB hematopoietic cells is dissimilar at various stages, the mechanism of which should be further studied.
