Effect of phytic acid on expressions of gene IkB-α and growth of gastric carcinoma cells

  • Objective To explore the effect of phytic acid on apoptosis of gastric carcinoma cells and mechanisms of proliferatory inhibition.Methods The inhibiting action of phytic acid on SGC-7901 cells were examined by MTT assay.The effect of IP6 on mor phology alterations of SG C-7901 cells by reverse discrepancy microscope observation and the effect on apoptosis of SGC-7901 cells was examined by Comet Assay,and the expressions of IkB-and NF-kBP65 protein were detected by Western blot method.Results From MTT assay,phytic acid was observed with the inhibition on the growth of gastric cells in dose and time dependent manners,the rate of cell growth suppression was 26.35%,53.20%,69.29%,86.65% and 93.61% at different concentrations,respectively.Cell growth of dosage group was inhibited.Rate of DNA tail of experimental groups was significantly higher than that of control group,and the rate of DNA damage was 5.9%,11.0%,34.6% and 63.1%.The expressions of IkB-α protein in phyt ic acid group were higher compared with control group,and increased in a dose-dependent manner.Expr essions of NF-kBP65 protein in phytic acid group was lower compared with control group,and reduced in a dose-dependent manner.Conclusion Phytic acid can inhibitit the proliferation of gastric carcinoma SGC-7901 cells which may be correlated to apoptosis associated gene IkB-α.
