Dynamic changes of cytokine signaling suppressor mRNA in rats with acute liver failure

ObjectiveTo observe the expression of cytokine signaling suppressors(SOCSs)mRNA in rats with acute liver failure(ALF).MethodsThe model of ALF was induced in rats by D-galactosamine/lipopolysaccharide(D-GalN/LPS).Then,the gene expressions of SOCS-1 and SOCS-3 in the liver was determined by RT-PCR,the TNF-α and IL-6 level was estimated by ELISA at 2 h,6 h,12 h,24 h and 48 h after the injection of D-GalN/LPS.ResultsThe TNF-α and IL-6 level were significantly elevated and reached the peak at 6th hour(P<0.01).Meanwhile,the gene expressions of SOCS-1 and SOCS-3 were obviously higher than those in the controls and reached the peak at 6th and 12th hour respectively after the injection of D-GalN/L PS(P<0.01).ConclusionAcute liver failure can induce the up-regulation of the gene expressions of SOCSs in rats,which closely correlates to the changes of TNF-α and IL-6,indicating that SOCSs may participate in regulating the balance of inflammatory response in acute liver failure.
