Effect of different ratios of n-6/n-3 PUFA on the expression of ER and p53 in MNU-induced breast cancer rats

Abstract
Objective To observe the effect of differen tratios of dietary n-6/n-3 poly unsaturated fatty acid on the expression of ER and P53 in the MNU-induced breast cancer rats.Methods Female SD rats were randomly divided into n-6 FUFA,10:1(n-6/n-3),5:1(n-6/n-3),1:1(n-6/n-3)and normal control groups.Except control group given physiological saline,all rats were given a singlipin jection of MNU50mg/(kg·bw)and switched to different diets containing fatty acids.After 8 weeks and 18 weeks,the expression of ER and p53 in the breast tissue of different groups were determ ined by RT-PCR and We stern blot.Results The expression of ER and p53 in breast cancer rats were higher than that of normal control(0.73±0.11,0.08±0.01)with the highest in n-6 group(1.32±0.18,1.43±0.56)and gradually decreased with the decreasing ratio of n-6/n-3.The expression of ER and p53 in 1:1 group(0.95±0.12,0.12±0.06)was significantly lower than that of in n-6 group.Conclusion Different dietary fatty acid compositions can diversely influence the expression of ER and p53 in MNU-induced breast cancer rats;1:1(n-6/n-3)can significantly inhibit the increasing of the expression of ER and p53 in breast cancerrats.
