Effect of luteolin and geraldol on redox potential in leukemia cell HL-60

Abstract
Objective To observe the effect of luteolin and geraldol on redox potential in leukemia cell HL-60.Methods The glutathion(GSH)and glutathione disulfide(GSSG)level were detected by O-phthalaldehy demethod.The changes of GSH/GSSG and the redox potential in HL-60 cells were analyzed.Futher more,we studied the effect with the pretreatment of N-acetylcy steine(NAC).Results After the treatment of 10 μmol/L luteolin or geraldol,the level of GSH was decreased;GSSG was increased;GSH/GSSG was decreased;the redox potential was increased.All the changes showed significant difference compared with the control at 2 hr or 4 hr after the treatment.The addition of NAC could prevent the reduction of GSH induced by luteo lin or geraldol,and partially suppressed their anti-proliferation effect on HL-60 cells.Conclusion Luteo lin and geraldol could significantly increase the redox potential in HL-60 cells,which may be an important mechanism for their anti-cance reffect.
