Abstract
ObjectiveTo investigate the β-amyloid precursor prote in(APP)gene changes induced by sodium sulfite.MethodsRT-PCR was used to study mRNA changes of APP and Western-blotting was used to detect the prote in level of APP in human embry onic kidney cell line 293(HEK 293)treated with sodium sulfite.ResultsRT-PCR results showed that treatment of 0.039-10 mmol/L Na2SO3 could not change the mRNA level of APP;Western-blotting results showed that 10 mmol/L Na2SO3 could decrease the protein level of APP in the cells significantly.On the other hand,0.039-2.5 mmol/L Na2SO3 could increase the prote in level of APP inside cells.Semi-quantity analysis showed that APP prote in levels were 1.596,1.630,1.556,and 1.446 times,respectively,when compared to that of the control.ConclusionNa2SO3 can cause significant increase of APP prote in level in HEK 293 and itmaybe related to Alzheimers' disease.But the change is not due to the change of mRNA of APP.
