Effect of Na-DMPS and NAC on brain energy metabolism in mouse with acute toxication of bromoxynil

  • ObjectiveTo investigate the effcet of sodiumidm ercaptopro pane sulfonate(Na-DMPS)and N-acetyl-L cysteine(NAC)on brain energy metabolism under acute toxication of bromoxnil in mouse.MethodsForty ICR mouse were randomly divided into five groups:control group,acute bromoxnil into xication(AI)group,Na-DMPS group,NAC group and NAC+Na-DMPS(NN)group.The contents of adenosine triphm sphate(ATP),adenosine diphosphate(ADP) and adenosine monophosphate(AMP)in the brain tissue were measured by RP-HPLC,and the values of energy charge (EC)were calculated.The activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in brain tissue were assayed with spectrophotom etric method.ResultsIn AI group,the contents of ATP,the levels of EC,the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase in the bra in tissue were significantly lower than that of in the control group(P<0.05).In Na-DMPS group,NAC group and NN group,the contents of ATP(1.53±0.38,1.81±0.41,2.04±0.38 μmol/g),the levels of EC (0.64±0.05,0.71±0.03,0.76±0.02),the activeties of Na+-K+-ATPa se(3.67±0.37,3.85±0.65,4.17±0.59 U/mg·prt)were higher than that of in AI group(P<0.05).ConclusionNa-DMPS and NAC and NAC alone can improve brain energy metabolism and increase the activities of Na+-K+-ATPase and Ca2+-Mg2+-ATPase under acute toxication of bromoxynil in mouse,and Na-DMPS in combination wtih NAC can achieve the best effect.
