Cloning and bioinformatics analysis of Ta 18 from Taenia asiatica oncosphere

  • ObjectiveTo analyze and predict the structural characteristics of Ta 18 gene from Oncosphere of Taenia asiatica cDNA club by bioinformatics.MethodsBy using the bioinformatics tools in bioinformatics webs site such as NCBI(http://www.ncbi.nlm.nih.gov/),ExPASY(http://www.expasy.org/),and other particular packages of biofomatic software as PCGENE and vector NTI suite 8.0,the Spef1-like gene and the coding region sequence from oncosphere of Taenia asiatica cDNA club was identified and analysed.Then the characteristics of structure and fuction of Ta 18 protein,including physical and chemical characteristics,modified sites and motifs in first structure,subcellular location,and secondary structure and potential epitopes were predicted.ResultsThe full cDNA sequence encoding Ta 18 includes a complete open reading frame of 396 bp in length,being a complete full-length gene encoding a putative protein of 131 amino acids.The protein presents 99% of identities with the 18kD gene of Taenia saginatus and 97% of identities with Taenia solium.The estimated molecular weight of Ta 18 is 14810.4 Da.The isoelectric point is 8.03,with stable phycical-chemical characteristics.There is a signal peptide and a structural domain of a functional domain of fibronectin type Ⅲ(FnIII).ConclusionTa 18 gene was screened from oncosphere of Taenia asiatica cDNA club and its structure and characteristics were predicted by bioinformatical method.
