Objective   					To explore genotoxic effects and possible mechanism of formaldehy de on the DNA crosslinks.
					   Methods   					V79 Chinese hamster cells were exposed to for maldehy de(FA)at different concentration of 75, 150, 300, 600, 1 200μmol/L and incubated for 1, 2, 4h, respectively.UV were used as standard DNA breaking agents, the DNA crosslinks in V79 Chinese hamster cells induced by different concentration of formaldehyde were detected with Single cell gel electrophoresis assay(SCGE, comet assay)in vitro.
					   Results   					Formaldehyde could cause DNA crosslinks of V79 Chinese hamster cells within the given doses.Except for the 75μmol/L dose, the crosslinks were increased with the concentration of formaldehyde and the exposed time in vitro.
					   Conclusion   					Formaldehyde was a DNA crosslinks agent that could cause DNA cross-links in V79 Chinese hamster cells, and the relationship between DNA crosslinks and concentration was a dose and time-dependent manner in vitro assay.
