Effect of grape procyanidin on proliferation of mice pancrease cell

  •   Objective   To study the effect of grape procyanidin (GPC)on the proliferation of normal and injured pancrease cell.
      Methods   (1) In vivo experiment, differet dose of GPC was given by mouth to the mice who radiated and not radiated by 60Co-γ-ray, then proliferation of pancreas cells was measured by MTT assay.(2)In vitro experiment, The pancreas cells of mice were pretreated and not pretreated by GPC and then were cultrured with diferent concentration of glucose and H2O2 respectively for 24 hours. The proliferation of normal and injured pancreas cell was determined by the same assay.
      Results   (1) Cell proliferation rate of radiation group and not radiation group given 150 mg/kg GPC were(3.16±0.13)% and(12.0±0.10)% respectively, higher than that of groups without GPC.(2)The cell proliferation of H2O2, 30 mmol/L glucose and 2.0 mmol/L H2O2 groups not pretreated by GPC were 0.120±0.008、0.070±0.002 and 0.063± 0.006 respectively, lower than that in groups pretreated by 100μg/ml GPC. All the differences above were significant in statistics.
      Conclusion   GPC can promote the proliferation of the normal pancreas cells and had protection on the proliferation of pancreas cells injured by radiation, glucose or H2O2.
