Genomic sequence determination and amino acid sequence deduction of dengue virus 2 type Mawei strain

  •   Objective   To analyze the partial gene sequence in NS1, E gene of dengue virus type 2 Mawei strain, and to deduce the amino acid sequence.
      Methods   Dengue virus type 2 Mawei strain and N GC strain were proliferated with C6/36 cells, respectively.The total viral RNA were extracted.The viral NS1 and E gene fragment were amplified with reverse transcriptase polymerase chain reaction(RT-PCR).Then the product of RT-PCR were sequenced, and the amino acid sequence was deduced by DNASIS software.
      Results   The E and NS1 gene of dengue virus type 2 Mawei strain were sequenced.Seven point mutations of NS1 gene and one base insertion of E gene were found, compared with DEN-2 NGC strain.This different sequence were accepted by GenBank.The accession number of NS1, E gene were A Y277402, A Y278226, respectively.
      Conclusion   The viral NS1 and E gene sequence were different between dengue virus type 2 Mawei strain and N GC strain.
