Study on oxidative damages and genotoxicity in mice induced by automobile exhaust

  •   Objective   To study the relationship between oxidative damages, and genotoxicity in mice by automobile exhaust.
      Methods   Mice were randomly divided into one control group and three exposure groups and were respectively exposed fresh air and different concentrations of automobile exhaust to hours each day for 5d in the device designed by investigator's laboratory.The concentrations of reactive oxygen species(ROS), malondialdehyde(MDA)and the activities of superoxide dismutase(SOD)in blood, liver, lung, testicle and micronucleus frequency in bone marrow cells were respectively determined.
      Results   The concentration of ROS in blood, liver, lung, testicle and MDA in lung, testicle of mice in automobile exhaust exposed groups were significantly higher than in negative control group(P < 0.01).Compared to negative control group, the micronucleus frequency in bone marrow polychromatic erythrocytes(PCE)in automobile exhaust exposed groups increased significantly(P < 0.01)and revealed dose effect relationship to a certain extent.There were correlative relationships between ROS in blood, liver, lung and the micronucleus frequency.
      Conclusion   Automobile exhausts could break the balance between oxidative system and anti oxidative system and damage chromosomes of cells.The cell oxidative damages induced by exposed automobile exhaust may play a key role in the effect of genotoxicity.
