Association of adipocytokine gene polymorphisms with type 2 diabetes mellitus: a meta-analysis

Abstract
Objective To evaluate the associations of gene polymorphisms in adipocytokines tumor necrosis factor α( TNFα),leptin receptor and adiponectin with type 2 diabetes mellitus( T2DM) using method of meta-analysis. Methods Hardy-Weinberg equilibrium( HWE) test was used to check genetic equilibrium between genotypes for the selected literature. A fixed or random effect model was used on the basis of heterogeneity. Publication bias was quantified and examined with the Begg’s and Egger’s method. The meta-analysis was performed with Review Manager 5. 1 Software. Results Fifty reports met the inclusion criteria for this meta-analysis. No significant publication bias was identified in the selected literature. There were no statistical significant results found regarding the association of leptin receptor( LEPR) single nucleotide polymorphism( SNP)-668A > G and T2DM. However,for TNFα SNP-308G > A,the odds ratioORand 95%confidence interval 95% CIfor A vs G allele and GA + AA vs GG were 1. 38( 1. 11- 1. 71) and 1. 26( 1. 05- 1. 51),respectively,with statistical significances. For adiponectin SNP + 45T > G,the OR( 95% CI) for G vs T allele and GG +TG vs TT was 1. 17( 1. 03- 1. 33) and 1. 16( 1. 01- 1. 34). Conclusion There is no significant association between the LEPR SNP-668A > G polymorphisms and risk for T2DM. However,TNFα SNP-308G > A and adiponectin SNP + 45T >G might be risk factors for type 2 diabetes mellitus.
