Effect of coal tar pitch smoke extracts on expression of inflammatory cytokines of THP-1 cells

Abstract
Objective To explore the effect of coal tar pitch smoke extracts( CTPE) on the exprission of inflammatory cytokines in monocyte-macrophages( THP-1). Methods THP-1 cells were treated with or without 2 μg / mL CTPE( dimethylsulfoxide as vehicle) for different time( 24,48,and 72 hours). The expression of interleukine 1-β( IL-1β),interleukine 8,IL-8) and tumor necrosis factor( TNF-α) mRNA were determined by real-time PCR. The release of TNF-αwas determined using enzyme-linked immunosorbent assay( ELISA). Results The expressions of TNF-α,IL-1β,and IL-8 mRNA of CTPE treatment groups were significantly different from those of the control group( P < 0. 05 for all). The expression level of IL-1β mRNA was 2. 22 ± 0. 33 after treated with 2 μg / mL CTPE for 24 hours,while the IL-8 and TNF-α mRNA level were 4. 64 ± 3. 03 and 3. 15 ± 0. 22 for 72 hours,all of which were higher than those of control and blank groups( P < 0. 05). The release of TNF-α for 72 hours treatment group was 538. 44 ± 53. 88 pg / mL,which was higher than that of other groups( P < 0. 05). Conclusion CTPE could promote THP-1 cells to express TNF-α,IL-1β,and IL-8.
