皮肤腐蚀性试验大鼠经皮电阻替代方法建立

Transcutaneous electrical resistance test as an alternative method of skin corrosion test in rat

  • 摘要: 目的 建立大鼠经皮电阻(TER)试验,并初步评价试验方法的可行性。方法 制作离体皮肤固定装置,用正常大鼠皮片固定于装置后的TER值变化及病理组织学变化来评价其可行性,参照OECD TG430的方法,采用10种化学物进行TER试验,并统计分析TER试验结果与动物试验结果的一致性。结果 正常离体皮肤固定装置24 h期间能维持屏障功能稳定,3次重复实验TER值变化曲线一致,24 h时皮片组织形态结构仍然完好;TER试验对10种化学物的腐蚀性判别结果与动物试验的结果具有一致性(Mcnemar,P=0.500;Kappa=0.615,P=0.035),灵敏度为100%,特异度为66.7%。结论 TER试验可替代家兔皮肤腐蚀性试验用于评价化学物的皮肤腐蚀性。

     

    Abstract: Objective To establisha method of transcutaneous electrical resistance(TER)test and to evaluate the feasibility of the method in rat.Methods TER value and histopa thological changes of normal rat skin discs were observed to evaluate the feasibility of the isolated skin fixing aPPARγatus assembled.Tenchemicals were tested according to the Organization of Economi Cooperation and Development Guide line for Testing of Chemicals No.430.Statistical analysis was conducted to determine the relevance of the TER test compared with the data of in vivotest.Results The barrier function of normal skin discs fixed in the aPPARγatus were stable with in 24 hours and the TER value variation curve swere repea table.The morphological structure of the skin discs was intact with in 24 hours.The TER test was concordant with in vivotest for the ten reference chemicals(Mcnemar,P=0.500;Kappa=0.615,P=0.035)with the sensitivity of 100% and specificity of 66.7%.Conclusion The isolated skin fixing aPPARγatus could be used for TER test.The TER test could a lternate skin corrosion test for testing corro sive chemicals.
