白藜芦醇对高脂饮食大鼠心肌PPARγ/AP-1信号影响

Effects of resvertrol on PPARγ/AP-1 in heart of rats with high-fat diet

  • 摘要: 目的 观察白藜芦醇对高脂饮食大鼠心肌过氧化物酶增殖物激活受体(PPARγ)及其下游信号分子激活蛋白-1(AP-1)表达影响。方法 雄性SD大鼠32只,随机分为对照、高脂、吡格列酮、白藜芦醇4组;分别喂饲普通、高脂饲料,灌胃给予吡格列酮、白藜芦醇10 mg/(kg·d),连续6周;分别检测心肌PPARγmRNA、PPARγ、AP-1蛋白表达变化。结果 高脂组PPARγmRNA表达(0.36±0.04)较对照组(0.48±0.04)明显降低(P<0.05);白藜芦醇组(0.54±0.11)较高脂组表达明显升高(P<0.05);高脂组PPARγ蛋白表达(0.40±0.02)较对照组(0.52±0.02)明显降低(P<0.05),而AP-1表达明显升高(P<0.05);白藜芦醇组PPARγ表达较高脂组升高,AP-1表达较高脂组降低(P<0.05)。结论 白藜芦醇对高脂状态下大鼠心肌PPARγ/AP-1信号转导通路有激活作用。

     

    Abstract: Objective To observe effects of resveratrol on the expression of peroxisome proliferator activated receptor (PPARγ)and its downstream signaling molecule activator protein 1(AP-1)in myocardia of rats with high fat diet.Methods Thirty two male Sprague-Dawley rats were randomly divided into control(C),high fat diet(HF),pioglitazone positive(PI),and resveratrol group(RESV)and were fed with normal and high fat diet,respectively.At the same time,PI group and RESV group were treated with 10mg/kg·d body weight pioglitazone,10mg/kg·d body weitgh resveratrol by gavage daily for 6 weeks.PPARγ mRNA was measured by reverse transcription PCR(RT-PCR).The expressions of PPARγ and AP-1 were detected by western blotting.Results Compared with group C(0.48±0.04),the expression of PPARγ mRNA in HF group(0.36±0.04)was lower(P<0.05).Compared with group HF,PPARγ mRNA in RESV group (0.54±0.11)was miproved(P<0.05).Compared with group C(0.52±0.02),the protein level in group HF(0.40±0.02) was reduced(P<0.05),whereas the level of AP-1 prote in in group HF was remarkably miproved(P<0.05).The level of PPARγ in RESV group was improved compared to HF group(P<0.05)and the level of AP-1 was reduced(P<0.05).Conclusion Resveratrol can activate PPARγ/AP-1 signaling pathway in myocardia in rats with high fat diet.
