溴氰菊酯对γ-GCS活力和谷胱甘肽含量影响

Effect of deltamethrin on gamma-glutamylcysteine synthetase activity and reduced glutathione content

  • 摘要: 目的 探讨溴氰菊酯(DM)对PC12细胞γ-谷氨酰半胱氨酸合成酶(γ-GCS)活力和谷胱甘肽(GSH)含量的影响。方法 用0、1、10、100μmol/L DM处理PC12细胞6或12 h后用四甲基偶氮噻唑蓝(MTT)法检测细胞毒性;用0、10、100μmol/L DM处理PC12细胞6或12 h后,超速离心分离细胞胞质碎片,邻苯二甲醛(OPA)柱前衍生高效液相色谱(HPLC)荧光法检测组织中γ-GCS酶活力和GSH含量。结果 100μmol/L DM处理细胞6、12 h的细胞存活率为(0.862±0.053)、(0.746±0.101),均低于对照组,差异均有统计学意义(P<0.01或P<0.001);与对照组γ-GCS酶活力(1.78±0.26)pmol/106细胞.min比较,10、100μmol/L DM处理细胞6 h组γ-GCS酶活力为(1.66±0.20)、(1.73±0.09)pmol/106细胞.min,差异均无统计学意义;与对照组GSH含量(10.18±0.23)pmol/106细胞比较,10、100μmol/L DM处理细胞6 h GSH含量为(10.19±1.18)、(10.26±0.29)pmol/106细胞,差异均无统计学意义;与对照组γ-GCS酶活力(1.94±0.19)pmol/106细胞.min和GSH含量(9.57±0.39)pmol/106细胞比较,10μmol/L DM处理细胞12 h组γ-GCS酶活力(1.73±0.21)pmol/106细胞.min和GSH含量(9.72±0.41)pmol/106细胞差异均无统计学意义,100μmol/L DM处理细胞12 h组γ-GCS酶活力(1.62±0.09)pmol/106细胞.min差异有统计学意义(P=0.006)。结论 较高浓度的DM处理细胞较长时间才抑制γ-GCS酶活力,可能与DM的细胞毒性作用有关。

     

    Abstract: Objective To explore the effect of deltamethrin(DM)on gamma-glutamylcysteine synthetase(γ-GCS) activity and reduced glutathione(GSH)content in PC12 cells.Methods PC12 cells were treated with 0,1,10,100 μmol/L DM for 6 hours or 12 hours,respectively.Cytotoxicity of DM on PC12 cells was measured by methyl thiazolyl tetrazolium(MTT)assay.The cytosolic fraction of PC12 cells was used to determine reduced glutathione(GSH)content andγ-GCS activity by reversed-phase high performance liquid chromatographic assay with o-phthalaldehyde pre-column derivatization.Results The survival rate of PC12 cells treated with 100μmol DM for 6 hours or 12 hours was lower than that of control group(P<0.01,P<0.001).In PC12 cells treated for 6 hours,there was no effect onγ-GCS activity and GSH content.In PC12 cells treated for 12 hours,there was no difference inγ-GCS activity and GSH content between low concentration and control group.Theγ-GCS activity of the high concentration group was significantly lower than that in the control group(P<0.05).Conclusion These findings demonstrate thatγ-GCS activity in PC12 cells could be inhibited by high concentration of DM treatment for a long time.
