血液中6种苯系物吹扫捕集/气相色谱/同位素内标质谱串联法测定

Determination of six benzene series in blood with purge-trapping/GC/MS and isotope internal standards series process

  • 摘要:
      目的  建立吹扫捕集/气相色谱/质谱联用技术同时测定生物样本血液中6种苯系物含量的方法。
      方法  优化吹扫捕集实验条件,富集和解析血液中6种苯系物,然后用气相色谱质谱联用方法,以同位素内标法选择特征离子对6种苯系物进行定性和定量分析。
      结果  通过优化吹扫捕集参数,实现了血液中苯、甲苯、乙苯、间/对二甲苯、邻二甲苯6种苯系物的同时富集、解析和准确测定;本方法检出限为0.002~0.011 μg/L,线性范围为0.043~10.99 μg/L;低、高浓度加标回收率分别为72.78 %~81.03 %、82.27 %~109.09 %;对同一样品重复进行6次分析,相对标准偏差(RSD)为3.47 %~7.36 %。
      结论  本研究所建立方法操作简便、周期短、分离度好,准确度和精密度均能满足分析测试要求,适用于生物样本血液中6种苯系物同时检测分析。

     

    Abstract:
      Objective   To establish an analytical method for the determination of 6 benzene series in whole blood.
      Methods   Benzene series compounds in whole blood were concentrated with purge and trap method and then 6 benzene series were measured quantitatively using gas chromatography/mass spectrometry with isotope internal standard process.
      Results   Benzene, methylbenzene, ethylbenzene, m-xylene p-xylene, and o-xylene in whole blood samples were concentrated and measured simultaneously after optimization of purge and trap parameters. For the method established, the detection limit was 0.002 – 0.011 μg/L; the linear range was 0.043 – 10.99 μg/L, and the recovery rates were 72.78% – 81.03% and 82.27% – 109.09% for low and high concentration samples. The relative standard deviation (RSD) ranged between 3.47% – 7.36% for 6 repeated determinations of the same sample.
      Conclusion   The established method is easy to operate and of short duration, good separation, and desirable accuracy and precision for simultaneous determination of 6 benzene series in whole blood.
