Association of ABCG2 gene polymorphism with uric acid level among gout patients in tropical areas

      目的  研究ABCG2基因多态性与热带地区痛风患者尿酸水平的关系,为痛风患者的诊治及健康预警提供依据。
      方法  选择海南省海口市人民医院2014年1月 — 2016年12月的海南本地痛风患者572例,按照血液尿酸检测水平分为高尿酸组和尿酸正常组,提取全血基因组DNA后委托公司进行ABCG2基因SNP位点rs2231142基因分型,对组间基因分型频率分别进行统计学分析。
      结果  纳入研究的572例患者中,男性痛风患者317例,占55.42 %,高尿酸患者占49.84 %,正常尿酸患者占50.16 %;女性痛风患者255例,占44.58 %,高尿酸患者占47.45 %,正常尿酸患者占52.55 %。rs2231142位点在海南本地人群中可检出CC、CA、AA三种基因型,其在女性中的检出率分别为51.37 %、36.08 %、12.55 %,在男性中的检出率分别为51.74 %、29.97 %、18.30 % ,男女性3种基因型分布频率差异无统计学意义(P > 0.05);3种基因型的分布频率在男、女性高尿酸组,男、女性低尿酸组,男性高、低尿酸组,女性高、低尿酸组间的分布频率及等位基因分布频率差异均无统计学意义。
      结论  rs2231142遗传多态性与热带地区痛风患者血尿酸水平无显著关联。

     

    Abstract:
      Objective  To investigate the association between adenosine triphosphate-binding cassette transporter G2 (ABCG2) gene polymorphism and uric acid level among gout patients in tropical areas and to provide evidences for the warning, diagnosis, and treatment of gout.
      Methods  Totally 572 gout patients (317 males and 255 females) living in Hainan province were selected at Haikou Municipal People’s Hospital from January 2014 to December 2016 and assigned into a hyperuricemia group and a normal uric acid group according to their blood uric acid level. Whole blood genomic DNA of the participants were extracted and single nucleotide polymorphisms (SNP) of ABCG2 gene rs2231142 were genotyped by a company entrusted. The genotype frequencies of the loci were analyzed with SPSS 19.0.
      Results  Of the male and female participants, 49.84% (159/317) and 47.47% (134/255) were detected with hyperuricemia, while 50.16% and 52.55% showed normal uric acid level. Genotypes of CA, CC, and AA of ABCG2 rs2231142 gene were detected in the participants, with the detection rates of 51.74%, 29.97%, and 18.30% among the males and 51.37%, 36.08%, and 12.55% among the females, respectively; there were no significant gender differences in the detection rate (P > 0.05). No significant differences were observed in genotype frequency distributions of ABCG2 rs2231142 loci and their allele between male and female participants with hyperuricemia and normal uric acid and between the participants with hyperuricemia and low uric acid level.
      Conclusion  The polymorphism of ABCG2 rs2231142 is not significantly correlated with blood uric acid among gout patients in tropical areas.
