一株产生聚-β-羟基烷酸细菌的筛选和鉴定

Isolation and Identification of Strain of the Bacterium Accumulated Poly-β-hytdroxyalkanoate

  • 摘要: 从北京近郊分离出一株能积累聚-β-羟基烷酸的菌株,经分类鉴定,确定其为Xanthobacter autotrophicus.并确定了其最适生长条件。提纯的聚-β-羟基烷酸,经HNMR证明此多聚体由聚-β-羟基丁酸盐和聚-β-羟基戊酸盐组成。在2%GH培养基中,两者的比例为1:2.6,聚-β-羟基烷酸的最高含量达62.70%(W/DW).

     

    Abstract: The bacterium of No.of strain accumulated poly-β hydroxyalkanoate(PHA)was isolated from soil in the suburb of Beijing,and was characterized and identified as Xanthobacter autotrophicus by comparing with the typical characters in Bergey's Manual of Determinative Bacteriology(ninth edition).The optimal condition of the culture was studied.By identifying purifed PHA by 1H-NMR,PHA was a composition of poly-β-hydroxybutyrate (PHB)and poly-β-hydroxyvalerate(PHV).The ratio of PHB to PHV is 1:216.The highest content of PHA was 62.70% (W/DW)by couturing the baeteria in 2% GH medium.
