溴氰菊酯对大鼠脑钙调蛋白含量和蛋白激酶活性的影响

Efffect of Deltamethrin on Calmodulin Content and PKA, PKC Activities in Rat Brain

  • 摘要: 为探讨溴氰菊酯(Delta methrin,DM)神经毒作用机理,采用竞争性固相酶联免疫法(ELISA)和放射性同位素法分别观察了DM对不同脑区钙调蛋白(Calmodulin,CaM)含量和蛋白激酶(Protein kinase)活性的影响.结果表明,大鼠经DM处理后,大脑和小脑CaM含量无明显变化,而大脑皮层和海马中PKA,PKC活性明显增强,同时,脑细胞膜和细胞浆中PKC活性亦明显升高.说明蛋白激酶活性增高使脑内蛋白质磷酸化作用增强可能是DM神经毒作用机理之一.

     

    Abstract: To make further understanding the mechanism of the neutotoxicocity of Deltamethrin(DM),Calmodulin(CaM)content and P KA,P KC activities were determined in different brain regions.The result illustrated;No obvious change of CaM content was observed in cerebral and cerebellum after DM treatment.The activities of P KA and P KC were significantly induced by DM.Activities of P KC in membrance and cytosol were also increased by DM.All the results demonstrated that the phosphorylation of brain proteins by P KA and P KC may by one of the mechanisms of the neurotoxicity of DM.
